Chromatography Series: Christmas Edition

The scent of Christmas is in the air – vanilla in rice pudding/Ris á la Mande, cinnamon in mulled wine/Glögg, and cloves in gingerbread. But behind these festive aromas lie complex chemical profiles that chromatography reveals with beautiful precision.

Vanilla – The Queen of Christmas

Is your vanilla extract real? HPLC on a C18 column (e.g., water-rich mobile phase with 1% butanol) cleanly separates vanillin from synthetic markers like ethyl vanillin and the troublesome coumarin in just a few minutes. Real Bourbon vanilla shows a rich profile with p-hydroxybenzaldehyde, vanillic acid, and p-hydroxybenzoic acid – compounds missing in artificial versions. GC-MS with headspace solid-phase microextraction (HS-SPME) adds the full aromatic picture: guaiacol, vanillyl alcohol, and anisyl derivatives that make real vanilla irresistible.

Cinnamon – Warmth in Every Peak

GC-MS is the gold standard for cinnamon essential oil. A single run on a non-polar or mid-polar column (e.g., TRB-5, DB-5 or equivalent) shows trans-cinnamaldehyde as the dominant peak (up to 87% in Ceylon cinnamon), followed by eugenol, linalool, and β-caryophyllene. These profiles instantly distinguish premium Ceylon cinnamon from cheaper Cassia (high coumarin) – important for both flavour and safety. And did you know that cinnamaldehyde has anti-bacterial and anti-fungal properties?
